Title : Screening of biomarkers for acute ischemic stroke using bioinformatics analysis

Abstract:

Acute Ischemic Stroke (AIS) is a life-threatening cerebrovascular illness. The cause of AIS is unknown, and treatment options are limited. The study is to find out which genes are expressed in AISs and use that information to guide future research and treatment. The raw datasets GSE195442, GSE95204, and GSE60319, which included AIS and matching superficial temporal artery (STA) samples, were downloaded from Gene Expression Omnibus. The limma package in R software was then used to find differentially expressed genes (DEGs). The DEGs were subjected to hierarchical clustering analysis with gplot2 in R. Database for Annotation and Visualization and Integrated Discovery (DAVID).GO functional enrichment can be done using bioinformatic web tools. KEGG called The Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses were performed using DAVID and gene set enrichment analysis separately.
The points where the results intersected were chosen as shared KEGG pathways.  Retrieving Genes using bionformic tools, PPI analysis for DEGs implicated using KEGG pathways was done and visualised with Cytoscape software. As a result, we now have genes called Pyk and Spas, which, when overexpressed, can cause AIS. Spastin (spas) is a AAA ATPase family member that assembles into hexamers and severs microtubules along their lengths.Tubulin glutamylation levels are required for the spas product's microtubule-binding and severing functions. Mitosis, axon transport, synapse development, dendritic arborization, organelle tubulation, and lipid droplet metabolism are all thought to be involved. Gene that codes for proteins. The gene encoding pyruvate kinase (PSK) is involved in muscle development, glycolysis, and glucose homeostasis. Pyruvate kinase activity is predicted to be enabled. Glucose homeostasis and sucrose response are both regulated by this protein. It's found in the P granule and the cytoplasm. It can be found in a variety of structures, including the circulatory system of adults. The current study discovered essential genes in the aetiology of AIS, which will bring fresh insight into AIS treatment.
