The effect of flurbiprofen on oxidative stress levels in the experimental animal model of the seizures

Abstract:

Background: Approximately 1% of the world’s population possesses the epilepsy disorder. The research for novel drugs is ongoing because of the common drugs' adverse side effects and drug resistance. Epileptic seizures can initiate inflammatory pathways in the brain, which can further increase oxidative stress. Regarding the interconnected causal relations between inflammation, oxidative stress, and seizures, Flurbiprofen (FLU) was proposed as a candidate drug due to its anti-inflammatory and free radical scavenging activities.
Methods: The rats were divided into four groups, each including five rats. Three rats' groups received Pentylenetetrazol dissolved in physiological saline, two of them after being introduced to 5 and 10 mg/kg of FLU. The control group received only vehicle. The rat's blood samples were collected after cessation of seizures. To evaluate the candidate drug effects on oxidative stress and thus seizure, Total Antioxidant Capacity (TAC) and Total Oxidative Status (TOS) assays were conducted, and the results were compared between the groups.
Results: As expected, FLU significantly increased TAC (P<0.05) and decreased TOS levels (P <0.05) when compared with PTZ group. These effects were even more prominent in higher doses of the drug.
Conclusion: The results suggest that FLU enhances protection against oxidative stress in the brain, thus possibly lowering the harm of acute seizures. This study was based on models of acute seizure; therefore, other studies are needed for the chronic models.
